I don't know anything about the comic books, but the game's explanation is that the infant Loki was orphaned after a battle and Odin took the child and raised him as his own son (and Thor's brother).

Umm I think you can kill him... not stab him persay (actually my brother says you can)... but you can kill him by making him drink waaay too much.
EDIT: i forgot he just passes out. but my brother reminded me that you can drop all of your items, stab him, go outside, find that guy, and come back in and he's dead
